rspo1 required development of gonads, regardless of sex. has been found in mice eleven days after fertilization. induce cell proliferation, acts synergistically wnt4. stabilize β-catenin, activates downstream targets. if both deficient in xy mice, there less expression of sry , reduction in amount of sox9. moreover, defects in vascularization found. these occurrences result in testicular hypoplasia. male female sex reversal, however, not occur because leydig cells remain normal. maintained steroidogenic cells, unrepressed.

ovaries

rspo1 necessary in female sex development. augments wnt/β catenin pathway oppose male sex development. in critical gonadal stages, between 6 , 9 weeks after fertilization, ovaries upregulate while testes downregulate it.

mucositis

oral mucosa has been identified target tissue rspo1. when administered normal mice, causes nuclear translocation of β-catenin region. modulation of wnt/β catenin pathway occurs through relief of dkk1 inhibition. occurrence results in increased basal cellularity, thickened mucosa, , elevated epithelial cell proliferation in tongue. rspo1 can therefore potentially aid in treatment of mucositis, characterized inflammation of oral cavity. unfortunate condition accompanies chemotherapy , radiation in cancer patients head , neck tumors. rspo1 has been shown promote gastrointestinal epithelial cell proliferation in mice.
